摘要:
通過微波消解法,原子吸收分光光度計(jì)對(duì)當(dāng)歸中鉛的吸光度進(jìn)行測(cè)定,采用Box-Behnken Design設(shè)計(jì)試驗(yàn)方法以確定最佳儀器分析條件;通過單因素試驗(yàn),選取灰化溫度、灰化時(shí)間、原子化溫度、原子化時(shí)間四因素進(jìn)行三水平試驗(yàn),通過Box-Behnken Design建立數(shù)學(xué)模型,方差分析、響應(yīng)曲面分析、愿望函數(shù)優(yōu)化尋求最佳條件;結(jié)果顯示建立的數(shù)學(xué)模型R2=0.9671,R2Adj =0.9342說明該模型具有很好的可信度,可用此模型進(jìn)行預(yù)測(cè),變異系數(shù) CV %=3.62 %,表明在該試驗(yàn)條件下,變異程度較小,該模型具有較好的重復(fù)性,當(dāng)灰化溫度為525℃,灰化時(shí)間為20s,原子化溫度為1550℃,原子化時(shí)間為5s時(shí)吸光度最大為0.24;因此,本研究可為原子吸收分光光度計(jì)測(cè)定鉛元素提供很好的方法理論基礎(chǔ)。

Abstract:
The absorbance of lead in flour was determined by microwave digestion method and atomic absorption spectrophotometer. Box-behnken Design was used to determine the best instrumental analysis conditions. Through the single-factor test, the four factors of ashing temperature, ashing time, atomization temperature and atomization time were selected for the three-level test. The mathematical model was established by Box-Behnken Design, and the optimal conditions were sought by variance analysis, response surface analysis and desire function optimization. Results show that the established mathematical model of the R2=0.9671,R2Adj =0.9342 shows that the model has good credibility, this model could be used to predict the coefficient of variation CV % = 3.08%, indicates that under the condition of the experiment, variation is small, the model has good repeatability, when ashing temperature is 525℃, ashing time is 20s, atomization temperature is 1550℃, atomization time for 5s absorbance maximum 0.24; Therefore, this study can provide a good theoretical basis for the determination of lead by atomic absorption spectrophotometer.
